Research Statement: Our research group studies the process of pattern formation. In particular we are focused on the Hedgehog signal transduction pathway and its regulation of cell fate specification. Our present goals are to understand how the signal is relayed from the cell surface to the transcription factors Ci in Drosophila and homologous Gli1, 2, and 3 transcription factors in mammals. We are pursuing three lines of research: In collaboration with Andreas Matouschek’s group, we are examining how the proteasome partially degrades Ci, Gli2 and Gli3. This process appears to depend upon the tightly folded zinc finger domain and a stretch of amino acids with low complexity. By exchanging domains and modifying their sequence and relative position, we hope to understand how partial proteolysis is accomplished. To identify additional components of the Hedgehog signal transduction cascade, an RNAi suppressor/enhancer screen has been carried out in a fu mutant background. To date we have screened kinases, phosphatases, acetylases, deacetylases, proteins that bind known components of the Hedgehog pathway and 1000 random genes. Roughly 50 genetic interactions were identified. The best candidates are being further characterized to identify which are directly involved in Hedgehog signaling and what aspects of the process they regulate. In collaboration with the Meade lab, we are testing a cobalt Schiff base that is specifically targeted to the Ci/Gli proteins for its ability to block the function of these transcription factors.
