Description

Objectives: To (i) identify enzymes that enable the production of structurally homogeneous glycoproteins, (ii) develop a generalized step-wise in vitro methodology for synthesis of glycans on demand, and (iii) prepare three glycoproteins: hIgG, hEPO and hBChE. Method: The proposed work combines (i) a cutting-edge cell-free protein synthesis system, (ii) newly established methods in prokaryotic glycosylation, and (iii) a high-throughput label-free screening tool to discover enzymes for the production of humanized glycoproteins.
