To work out how the biochemical activities that CSH characterized in the papillomavirus E1 protein affects the in vivo functions of E1. I will utilize our expertise in analyzing viral DNA replication of the HPV 31 genome in human foreskin keratinocytes during the latent and productive stages of the viral life cycle. I will test the E1 mutants that affect phosphorylation sites and protein-protein interaction sites. I will also provide samples from differentiated and undifferentiated HFKs for analysis of expression levels of cellular proteins that may be involved in regulation of the viral DNA replication.
