Bridge Funding Request for Project 1 (Surmeier): Our goal during the bridging period is to complete our studies focused on the disease-modifying potential of selective inhibition of Cav1.3 channels in substantia nigra pars compacta (SNc) dopaminergic neurons. These studies are directly linked to the interpretation of the on-going Phase 3 clinical trial with isradipine being sponsored by NINDS and to the development of complementary therapeutic strategies for disease modification. Bridge Funding Request for Project 3 (Bevan) Our goal during the proposed bridge funding period is to complete our Udall Center-funded studies focused on the pathological synchronization of the subthalamic nucleus (STN) by the motor cortex and external globus pallidus (GPe) in mouse models of Parkinson’s disease (PD). These studies are critical for understanding 1) the mechanisms underlying the emergence of abnormally correlated and coherent activity of the STN in PD 2) whether this abnormal activity pattern directly contributes directly to Parkinsonian motor dysfunction 3) whether correction/interruption of abnormal STN activity ameliorates Parkinsonian motor dysfunction.
