Description

The proposed research will use the technique of somatic brain transgenesis, injecting adeno-associated virus (AAV) into the ventricles of mouse pups on the day of birth, a procedure that my lab has used and published on before. In addition, it requires the use of the 5XFAD Alzheimer’s disease mouse model of amyloid pathology and neurodegeneration, which was generated in my lab. 1) My lab will breed the 5XFAD mice needed for this project, and do the AAV injections of the pups, using the AAV provided by Dr. Cepko. 2) We will then age these animals to the desired timepoints, and harvest their brains. One fixed hemibrain will be provided to Dr. Cepko’s laboratory for analysis of neuronal survival. Our lab will section and analyze the other hemibrain for Nrf2/GFP expression, amyloid pathology, neuroinflammation, and apoptosis using immunofluorescence/immunohistochemistry and widefield and/or confocal microscopy.
