Abstract

The assay development for detection and quantitation of fatty acid acyl conjugates of hydrocortisone (HC) is discussed. The gas chromatography-electron capture negative ion mass spectrometry (GC-ECNI-MS) techniques are used for the purpose. Fatty acid acylation of HC would alter the partitioning properties of the glucocorticoid making it more lipid-soluble and more likely to be cell-associated. Deacylation of the fatty acid conjugates of HC would also provide a mechanism through which the biological effect of the endogenous steroid may be prolonged. The sequence of fatty acid acylation and deacylation of HC in vivo may represent a mechanism through which HC modulates inflammation.
