Abstract

The differential protein expression was studied focusing on down regulation in the cytosol fraction of a breast cancer MCF-7 cell line selected for resistance toward the alkylating agent melphalan. A combination of cell fractionation, two dimensional gel electrophoresis, mass spectrometry and bioinformatics was used during the study. MCF-7 control and melphalan resistant MCF-7 cells were grown to confluence in serum supplemented media. Comparative image analysis of the protein patterns in the cytosol fraction of MCF-7 control and MelR MCF-7 revealed several proteins whose expression was altered between the two cell lines.
