The most rigorous definition of a top-down experiment involves high-resolution measurement of an intact molecular weight value and direct fragmentation of protein ions in the gas phase. Neil L. Kelleher at the University of Illinois at Urbana-Champaign describes how this technique turns MS data from whole and fragmented proteins into biological information-and the challenges that still remain.
