Abstract
The dopamine D2 receptor (D2R) plays an important role in mesencephalic dopaminergic neuronal development, and we have previously reported that Wnt5a interacts with D2R to promote the differentiation of dopaminergic neurons. Co-immunoprecipitation of Wnt5a and D2R, displacement of the D2R receptor antagonist [ 3 H]spiperone from D2R by Wnt5a, as well as glutathione S-transferase (GST) pulldown assays demonstrated that Wnt5a binds D2R. In this chapter, we present the co-immunoprecipitation, pull-down, and competition-binding assays used in the analysis of the D2R-Wnt5a interaction.
